Application of Three-Dimentional Mixture Gel in Cell Migration Research

摘要:

细胞迁移的研究中可用到的培养基质远不能满足需要.利用鼠尾胶和基底膜胶质(Matrigel)混合形成三维Collagen-Matrigel混合胶,该混合胶在培养液∶鼠尾胶∶基底膜胶质为1∶5∶4配比下,0.5 h内凝固满足组织培养要求.同时该混合胶经固定后在光学显微镜下结构维持良好,无颗粒.弥散性因子表达细胞团块和上菱唇组织块共培养于该三维Collagen-Matrigel混合胶培养基质中,结果显示细胞迁移良好,SDF-1因子和Slit因子分别对小脑上菱唇细胞具有化学吸引作用和化学排斥作用.

Abstract:

The matrix used in cell culture cannot match the requirement during the cell migration research in vitro at present. The three-dimentional collagen-matrigel mixture gel was studied which was made up of culture medium∶collagen∶matrigel by different ratios. The mixture gel got solidified within half an hour at room temperature, and did not show visible particles under microscope observation after fixation. The results from co-culture of cell aggregate and upper rhombic lip (URL) explant indicated that SDF-1 has the chemoattraction to URL cells, while slit has the chemorepulsion to them. 
